Peptide Stacking: What Published Research Shows About Combinations

Peptide stacking and combination protocols from published research. Which peptides are studied together, rationale for stacking and what data exists.

Peptide Stacking: What Published Research Shows About Combinations

Combining multiple peptides in a single experiment, referred to as peptide stacking, is a popular subject in peptide studies. Though the idea is usually based on complementary mechanisms, there is in fact very little evidence supporting the vast majority of combinations. In this article we will look through the evidence of different stacks to differentiate those based on strong scientific logic from those based only on assumption.

What Peptide Stacking Means in Research

Complementary Mechanisms

The concept of stacking is easy to understand: Peptide A is designed to act on the X pathway and Peptide B on pathway Y and their combination results in a better or larger effect than any of them separately. This works well provided the pathways are truly independent and do not hinder each other. A good example of such a situation is BPC-157 and TB-500 as the two have different mechanisms although they both relate to tissue healing.

Additive vs Synergistic Effects

To be additive means that the result is equal to the simple addition of its parts: i.e. 1 + 1 = 2. Synergy would be the opposite: it implies that the actual effect is more than just the sum of the additions: e.g. 1 + 1 > 2. In the majority of cases we assume that there is synergy in peptide stacking. However, one cannot prove the fact of synergy without carrying out a special kind of experiment aimed at testing every ingredient separately from each other and then applying the very combination in order to prove that its final effect exceeds the simple addition of its parts. Up to now, no experimental study has been published proving synergy in the case of any combinations of researched peptides. So at best, we can say that the evidence tends to support additive effects based on the fact that the mechanisms of each ingredient can be complementary to each other.

BPC-157 + TB-500: The Most Discussed Stack

Published Data on Each Compound

BPC-157 can boast more than 100 studies that demonstrate its ability to improve healing via VEGF enhancement, nitric oxide regulation, and FAK-paxillin activity. TB-500 (Thymosin Beta-4) is backed by more than 50 studies highlighting its healing capabilities via G-actin isolation and effects on migration. Both compounds have compelling independent scientific evidence behind them for healing purposes. They are not only from different worlds, but their mechanisms do not coincide.

Rationale for Combining

BPC-157 makes a favorable biochemical milieu at the site of trauma, characterized by the presence of greater amounts of growth factors, increased angiogenesis, and reduction in excessive inflammation. TB-500 is responsible for physically motivating the cells involved in repair (i.e., fibroblasts, endothelial cells, and keratinocytes) to move into the damaged site. The idea is that both the chemical and cellular components necessary to implement the healing process are in place. The reasoning seems to be sound but is not backed by controlled studies.

Pre-Blended vs Separate Vials

PeptideGod's price for pre-mixed BPC-157 + TB-500 blend is $59.95. Meanwhile, if you buy vials separately, the total cost for BPC-157, which is $34.95, and TB-500, which is $35.00, is $69.95. So, it can be seen that by using the blend, you are saving about $10 and removing the inconvenience of having to draw from two different vials. However, with ready-for-use vials, there is a compromise in that one does not have the freedom to vary the ratio between the two peptides and so does not have the luxury of adjusting the dose of each compound independently.

Ipamorelin + Tesamorelin

GHRP + GHRH Rationale

The growth hormone secretagogue Ipamorelin functions by making use of the ghrelin receptor (GHSR) while Tesamorelin is a growth hormone-releasing hormone analogue that works on the growth hormone-releasing hormone receptor. These represent two separate input channels for the release of growth hormone from the pituitary. The hypothesis states that simultaneous stimulation of the two pathways provides increased GH release than activation of either pathway by itself. Several published studies concerning combinations of GHRP and GHRH support the idea of increased GH pulse amplitude due to activation of both receptors.

Available Combined Products

The product of Tesamorelin and Ipamorelin by Real Peptides is priced at $43.20, and comes with the COA documentation as verification. If one buys standalone Tesamorelin for $45.60 and standalone Ipamorelin from PeptideGod for $39.95, he/she would end up paying $85.55 to two different vendors. So it is evident that the Real Peptides product is cheaper and meets all documentation requirements with just one vendor.

Semax + Selank

Nootropic + Anxiolytic

Semax aims for cognitive enhancement and BDNF upregulation, while Selank focuses on anxiolytic GABA-ergic pathways. Together they help improve both cognitive functions and anxiety as both areas of brain functioning tend to intersect. Stress-effects on cognitive functioning are distinguishable from cognitive performance in a state of relaxation; this justifies the combination of two different compounds.

Nasal Co-Administration

Semax and Selank function as nasal spray products, meaning they can be combined in a very easy manner. PeptideGod has a pre-mixed nasal spray with both Semax and Selank available for $59.95, while also selling the standalone product NA-Semax-Amidate for only 49.95. The product is similar to combination shown for BPC-157 and TB-500 in that it eliminates any possibility of modifying the ratios of the active agents in the solution.

Stacks Without Published Combination Data

Why Absence of Data Matters

Some online forums suggest mixes such as BPC-157 + Ipamorelin, BPC-157 + MOTS-c, or "complete stacks" containing 3+ peptides. There are no published studies regarding efficacy and safety of these combinations. The absence of data doesn't mean the combination can be harmful or unsafe, but it means that there is no reliable information. There is always a risk of negative interaction between two compounds that have not been previously studied together. Two peptides that are safe on their own can play together in a way that produces unexpected results due to the fact that they may inhibit or interfere with each other’s mechanisms of action. Professor Oh suggests that it may not be a big problem for drugs that were previously thoroughly studied, but nothing is known about risky drugs.

FAQ, Peptide Stacking

Is it safe to combine peptides?

No studies analyzing the safety of combinations of different research peptides have been published. Each of the individual compounds (i.e. BPC-157, TB-500, Ipamorelin, Semax) is known to perform well in terms of safety in studies currently available. Theoretical risks associated with combining such compounds are low, yet "low risk in theory" does not equate to saying that any such combination is "safe." So all combinations remain unproven, and they exist between the evidence and practice gap.

Which peptide stacks have published research?

To speak strictly, none of the specific combinations of products offered by vendors have been tested in scientific studies. The evidence closest is obtained for the use of GHRP + GHRH (a general class including Ipamorelin + Tesamorelin) – there is published evidence showing that there is a stimulated GH release due to dual receptor stimulation. The BPC-157 + TB-500 combination has a good mechanistic basis while failing to have its combination studied. Semax + Selank has complementary mechanisms but no studies have been published regarding the combination studied.

Is a pre-blended vial better than mixing separately?

Vials that are pre-blended are cheaper and more convenient (one reconstitution, one draw). Separate vials provide you with drug dose control, you can adjust your quantity of each peptide as you want. If the predetermined ratio that is contained in the blend is what is in accordance with your protocol, then the blend will work better for you in terms of the cost. If you need a different ratio or want to use only one compound on specific days, then you can benefit more from the use of separate vials instead.
